TERT基因位于染色体5p15 33上,包含16个外显子和15个内含子,编码 端粒酶 逆转录酶,在保持端粒稳定、基因组完整、细胞长期活性和肿瘤细胞无限增殖能力等方面具有重要作用 [7]。 它与端粒酶RNA组分(TERC)共同组成端粒酶,是一种维持细胞内端粒长度的重要物质,因此,它也决定着端粒酶的活性。 在正常细胞中,端粒酶活性通常被严格控制,在大多数组织中水平很低,仅在生殖细胞和造血干细胞等少数细胞中存在并受到高度调控。 而对于肿瘤细胞,超过90%都表现出端粒酶过度表达,从而可以维持或延长端粒的长度,并导致细胞的恶性增殖及永生 [8]。 TERT的激活主要是由于启动子区发生了基因突变,最常见的就是TERT转录起始位点上游124bp的C228T点突变和146bp的C250T点突变。
